    Individual cellular clocks in the suprachiasmatic nucleus (SCN), the circadian center, are integrated into a stable and robust pacemaker with a period length of about 24 hours. We used real-time analysis of gene expression to show synchronized rhythms of clock gene transcription across hundreds of neurons within the mammalian SCN in organotypic slice culture. Differentially phased neuronal clocks are topographically arranged across the SCN. A protein synthesis inhibitor set all cell clocks to the same initial phase and, after withdrawal, intrinsic interactions among cell clocks reestablished the stable program of gene expression across the assemblage. Na+-dependent action potentials contributed to establishing cellular synchrony and maintaining spontaneous oscillation across the SCN.

    It is well known that the function of the mammalian circadian clock is altered by aging. In this study, we investigated whether a systemic pro-aging factor, beta2-microglobulin is involved in aging-associated alternation of the circadian clock function in mice. We found that the administration of beta2-microglobulin induced the elongation of the period in the free-running rhythm of clock gene expression in the cultured suprachiasmatic nucleus. It also reduced the density of arginin-positive neurons and astrocytes in the suprachiasmatic nucleus. These results suggest that a systemic pro-aging factor, beta2-microglobulin is involved in aging-associated alternation of the circadian clock function in mice.

    In the hippacampus, it was reported that the cell division occurs during specific time-window in a day. However, its mechanism remains to be fully clarified. In this study, we tried to clarify how the molecular clock regulates the circadian proliferation of the neural stem cells. We found that Sirt1 is causally involved in the circaidan proliferation of the neural stem cells using automatic real-time monitoring system for cell division.

    We developed ultra-stable nanoparticles of CdSe conjugated with anti-Her2 antibody as a novel functional fluorescent probe for in vivo imaging. After intravenous injection of this nanoparticle, we were able to detect the fluorescent signals and image of human esophageal cancer in tumor-bearing nude mouse from outside the body successfully. Furthermore, we developed new surgical system for endoscopic fluorescent surgery based on clinical application. Using this fluorescent detection system and CdSe as a nano-tracer, we succeeded in real time imaging of lymph networks and sentinel lymph node biopsy of gastrointestinal tract in pig model.

    Nanoparticles in appropriate size can be passively transported in malignant tissue. We performed in vivo imaging of the dynamics of nanoparticles in various sizes. Nanoparticles of 200nm did not leak in cancer tissue. In contrast, nanoparticles of 20, 40 and 100nm could be transported in cancer tissue. We also performed fluorescence nanoimaging of gastric sentinel lymph node of pigs. And we specified the first area for lymph node metastasis in a sentinel lymph node. These techniques are also utilized for ultrafine cancer operation.

    We have studied on the development of tomographic imaging technique of fluorescence in biological tissue for assays of biological function. Ultrasonic modulation of light based on the acousto-optic effect (so called ultrasound 'tagging') is applied for imaging of fluorescence distribution in the light-scattering media. Sound-field characteristics that affect the light by modulating its amplitude through variation of the refractive index in the medium were determined. With using focused ultrasound, selectively modulated fluorescence on a depth-axis of the medium can be detected. Ultrasound tagging technique applied measuring the optical absorption in light scattering media is well known, and it is principally based on the modulation of speckle pattern. On the contrary, in the case of fluorescence, displacement of scattering particles and variation of the refractive index that is induced by density distribution in a sound field might produce the intensity modulation of scattered light. We have experimentally shown that ultrasound tagging technique is also available for fluorescence measurement. In this paper, we demonstrate the result of tomographic images of fluorescence in dense scattering media using biological tissue, porcine muscle and bovine adipose. Tissue samples had the dimension of 40 × 40 mm in section and fluorescent region which had the 3mm size was embedded in the center of the tissue. The image of the fluorophore was determined with the spatial resolution of focus size of the ultrasound, suggesting the applicability of this technique for visualization of fluorescent probes in deep portion of living body for clinical application. We continue the research aiming to development of a novel diagnostic imaging system.

    CdSeナノクリスタルは、安定した作成法を確立したが、官能基を結合させるための外殻ポリマーコーティングが必要である。生体に無毒なシリカでコーティングを行ったところ、蛍光特性が減弱する現象が認められ、そのままでは本実験に使用できなかったため、種々の条件下で、改良したコーティング法を試行したが、その解決には至らなかった。

    Her2抗体を付加したCdSeナノクリスタルは坦癌マウス腫瘍特異的に集積し、体外からのイメージングができたが、高感度CCDカメラを組み込んだ装置と自家蛍光のsubtraction処理が必要である。800nmの蛍光で、約1cmの深部まで観察可能であった。
    (大動物によるセンチネルノードナビゲーション内視鏡下手術応用への基礎的実験)
    胸腔や腹腔など体腔内は皮膚の自家蛍光の影響が少なく、内視鏡下操作であれば基本的に暗い空間であるので蛍光波長を捉えるのに都合がよい。ブタを用いて大動物体内でCdSeナノクリスタルの蛍光検出を試みた。消化管内視鏡下に食道粘膜下層にCdSeを局注した場合の食道周囲リンパ節、反回神経周囲リンパ節を胸腔鏡で、胃粘膜下層に局注した場合の左胃動脈周囲リンパ節を腹腔鏡で観察した。このとき使用した体腔鏡は通常の手術用硬性内視鏡の先端にレーザー照射装置を組み込んだ自作のものを用い、ナノクリスタルは蛍光が長波長の800nmを用いた。リンパ節の蛍光が検出でき、少なくとも数十分観察が可能であることを確認した。センチネルナビゲーションに応用可能であり、官能基を付加したクリスタルであれば機能を持たせて、転移リンパ節だけに集積させることも可能となる。

    We have studied the monitoring technique of gene expression occurred in the internal body of mice. Optical methods to resolve the distribution of fluorophors expressed as gene reporters or exogenously administered fluorescent markers are investigated. In order to detect the fluorescence with spatial resolution, we have applied the coherent detection imaging technique, and also examined to apply a fluorescence life-time imaging and a spectral imaging technique to eliminate the auto-fluorescence, which is the background emission of tissue being a limiting factor to determine the minimum detectable signal The coherent detection imaging technique offers the wide dynamic range of optical detection and antenna properties to extract the straight forward scattering light in turbid media. We have combined this technique to apply for the detection of fluorescence in the biological tissue, and achieved the visualization of fluorophores embedded in the tissue at the depth up to 20mm from the surface. We conclude the potential usefulness of this technique to identify gene expressions originating from fluorescent proteins in vivo and this technique can also be applicable for a variety of biological sensing using fluorescent labeling technique.

    生体時計の分子機構の総括班会議を行った。本年度は特定領域研究の最終年度に相当するので、それぞれの班の研究代表者による4年間の研究成果の発表を行い、それに対する質疑応答を行った。本間班ではフィードバックループの形成に重要であるDec1,Dec2遺伝子を同定し、これがPer遺伝子に対する転写抑制作用が強いことを明らかにし、その成果はNatureに掲載された。また、clock変異体マウスを用いた研究から、視交叉上核内細胞間のカップリングが発振に垂要であることを明らかにした。柴田班では毎日一定時刻に餌の提示を行うことにより、末梢時計がリセットされることを世界に先駆けて発表した。グルコースの一過性の上昇が必要である可能性については深田班の研究で明らかにされた。深田班は体内時計の発振、リセット機構にMAPキナーゼのリン酸化が関与していることを、ヒヨコの松果体やマウスの視交叉上核、さらに培養細胞系で明らかにしてきた。福永班では、時計のリセット機構にCaMKII,IVがかかわっていることを明らかにした。また、共同研究の滝口らのグルーブが肝臓でプラスミノーゲン遺伝子発現に独自のリズム機構が働いていることを明確にした。海老沢班ではヒトのサーカディアンリズム障害と時計遺伝子多型について調べた。Per3遺伝子の変異と、睡眠リズム位相後退や非24時間リズム障害との連鎖が報告された。また、Per2蛋白のリン酸化にかかわっているカゼインカイネース1イプシロンの変異と、睡眠リズム疾患との関連性も述べられた。近藤班ではシアノバクテリアを用いた時計の分子機構が精力的に進められ、特にkaiA,B,C遺伝子産物のリン酸化や、多量体形成機構についての研究が進んでいた。評価委員の井上、永井各氏は研究成果の内容等に高い評価を下した。また、班員間の共同研究も十分行われているという評価を得た。
